Why does rofecoxib increase the probability of stroke?



Answer:
rofecoxib is one of the selective COX-II inhibitors this means that it inhibits COX-II enzyme within the endothelial cells of blood vessel &and not COX-I of platelets . so the prostaglandins produced by endothelial cells <which are responsible for the anti-platelet aggregation action>will shrink wherease the TXA2 produced by the platelets<which are responsible for platelet aggregation> are not affected. the lattice result is imbalance between TXA2 &PG so the probability of incidence of stroke will increases according to the artificial area.
Probably, I instinctively think it is the Result of Using a COX-2 Inhibitor Alone (TXA-2 May Not Be Lowered By Inhibition of this Iso-Enzyme Alone).
