I'm work in blood bank i ask what is the best way to separate platelets in units to give it to patient?

Wow... I would hope that since you work in a blood bank, you would be the one answering that question, not asking it.
centrifuge. platelets have less mass than red blood cells and will seperate from them in a centrifuge (red blood cells will go to the outside)
I would have thought centrifuging of blood for fresh platelet transfusion in thrombocytopenia or large volume transfusion.Your consultant haematologist will advise on protocols
